The New Zealand-based media group's acquisition of Yahoo's digital content businesses, which includes MySpace and its brand of similarly designed social networks, is expected to be completed in the next three to six months.
The exact stake in MySpace is not yet known, but a source with knowledge of the matter suggested it could be between 20 per cent and 25 per cent.
Yahoo declined to comment on the potential stake deal. MediaWorks also declined to comment.
MySpace's estimated $1.1bn (£608m) price tag is less than $300m higher than the $8.6bn buyout offer made by the Blackstone Group in August.
Last month, MySpace - which began as a college project by two students and has since evolved into a place to share a common life online - raised $300m in fresh capital to maintain its low-cost, high-growth model.
MediaWorks will also launch a new MySpace in New Zealand as part of its business, bringing the two regions closer together.
"Yahoo's MySpace creates an opportunity for MediaWorks to expand into the New Zealand market, where we currently have very little presence," said media group chief executive Tom Mockridge.
